AR Synthetic Response Element Construct (Androgen Receptor)

HT1080 fibrosarcoma cells were co-transfected with the AR_synthRE (S900012) reporter construct and an AR cDNA construct according to SwitchGear's standard protocols. The transfected cells were then treated with 10nM R1881 or vehicle only (untreated) for 24 hours.

CREB Synthetic Response Element Construct

HT1080 fibrosarcoma cells were transfected with the CREB_synthRE (S900013) reporter construct according to SwitchGear's standard protocols. The transfected cells were then treated with 100nM PMA or vehicle only (untreated) for 4 hours.

ER Synthetic Response Element Construct (Estrogen Receptor)

HT1080 fibrosarcoma cells were co-transfected with the ER_synthRE (S900014) reporter construct and an ER cDNA construct according to SwitchGear's standard protocols. The transfected cells were then treated with 10nM beta-estradiol or vehicle only (untreated) for 24 hours.

GR Synthetic Response Element Construct (Glucocorticoid Receptor)

HT1080 fibrosarcoma cells were transfected with the GR_synthRE (S900015) reporter construct according to SwitchGear's standard protocols. The transfected cells were then treated with 100nM dexamethasone or vehicle only (untreated) for 4 hours.

HIF1a Synthetic Response Element Construct (Hypoxia)

HT1080 fibrosarcoma cells were transfected with the HIF1a_synthRE (S900016) reporter construct according to SwitchGear's standard protocols. The transfected cells were then incubated in a low oxygen condition (1% O2) or under normal conditions (untreated) for 24 hours.

HSF Synthetic Response Element Construct (Heat Shock)

HT1080 fibrosarcoma cells were transfected with the HSF_synthRE (S900017) reporter construct according to SwitchGear's standard protocols. The transfected cells were then incubated at 37 degrees C or 43 degrees C for 8 hours.

NFkB Synthetic Response Element Construct (Inflammation)

HT1080 fibrosarcoma cells were transfected with the NFkB_synthRE (S900018) reporter construct according to SwitchGear's standard protocols. The transfected cells were then treated with 50 ng/mL TNF-alpha or vehicle only (untreated) for 8 hours.

STAT1 Synthetic Response Element Construct (Inflammation)

HT1080 fibrosarcoma cells were transfected with the STAT1_synthRE (S900019) reporter construct according to SwitchGear's standard protocols. The transfected cells were then treated with 500 u/mL Interferon-alpha or vehicle only (untreated) for 8 hours.
